The objective of this study is to clarify the contribution of phospholipase A2 (PLA2) to airway inflammation in aspirin-sensitive (Asp(-)) asthmatic patients. We measured eicosanoid and PLA2 activity in sputum after aspirin challenge using a non-radioactive method. Sputum supernatant (SSN) of Asp(-) asthmatics but not of the non-aspirin-sensitive (Asp(+)) asthmatics has eicosanoid-producing activity and PLA2 activity. The release of arachidonate metabolites from eosinophils is mainly mediated by cytosolic phospholipase A2 (cPLA2), but that from basophils by secretory phospholipase A2 (sPLA2). The release of arachidonate metabolites from basophils is greater in Asp(-) than in Asp(+) asthmatics. In addition, the release of arachidonate metabolites from eosinophils and basophils in SSN is greater in Asp(-) asthmatics than in Asp(+) asthmatics. The amount of PLA2 activity in sp
